It is very difficult to say how long it will take to get a reply as it varies considerably from celeb to celeb. Being as you are in the UK, a first class stamp (30p) on your letter and another 30p on the self-addressed envelope (for the reply) should be plenty. I would suggest sending an A4 envelope for the reply - so it can take up to a 10x8.
It is difficult too to say whether you will get an actual autograph or a preprint / autopen. From experience of writing to Markham & Froggatt it appears that the autographs from their UK clients are genuine (I have got successes from Bill Nighy and just the other day from Sir Ian Holm). However, I have also approached them for autographs for about 10 of their US clients and I have had nothing back (after 6 months). The only US celeb I have got anything back from Markham & Froggatt was Patricia Arquette, and that was an autopen.
If you do get a genuine autograph back however, you must be prepared to wait anything up to 6 months or longer
