I would suggest leaving after the final curtain (after everyone takes their bow) Depends on where your seats are, how many people you see getting up and leaving, and how long you actually want to stand around the door and wait for the actors to come out. You could have a short wait or a super long one. The longest wait I had was 5 hours. The shortest was about an hour and a half.
Edited to add: And even standing by the stage doors is NOT a guarantee that you'll get close enough or even get an autograph from the actors. There is more than one way out of most of the theatres in NYC.
