How long should a person wait before sending out a 2nd request?

I have started sending out letters since April 2008, send out 500 letters & have about 100 autographed items in.

I am now working on 2 other fundraisers.

I am seeing people get their requests back faster than I have. Some in 2 weeks when I have sent out for the same person 4 months ago.

I am now working on autographed items to come in for the Rotary Auction that helps buy "TOYS for TOTS" in October.

Hats off to you, thats a great response rate you have there. 1 in 5 is not bad, and you will get more coming in in the coming months. Sadly, you often see replies for celebs you have written to, its just one of these things. Its up to you when to write again, or indeed IF you should write again. Its for charity, so I can't see you having much to lose, a LOT of celebs have consistently supported me over the years.
