It's hard to say, but i'm sure most wouldn't mind. In fact I doubt most would even remember that you had already sent a letter, considering how much fan mail they get (depending on how popular the person you're sending to).
But I would probably wait a while before sending your second, just to be sure

But yes, I am sure that Richard Griffiths, and most other people would be completely fine with signing your second item, provided you don't send it too close to your 1st, and cover the return postage
